Ongoing Research

1. The development of multi-omic single cell tools to investigate genomic editing in primary human cells. This work is rooted in developing new ways to scale a plate-based multi-omic assay with advanced microfluidics and robotics. The development of a robust and democratized method for investigation of CRISPR editing in primary human cells will unlock new avenues of study to greater our understanding of auto-inflammatory diseases

2. Understanding the genetic contribution of autoimmune variants on human B cell activation. This works involves the development of genetic editing approaches in primary human B cells and large scale screening techniques to unbiasedly discovers the genetic regulatory pathways in human B cell activation.

3. Using genetic editing tools to probe the coding and non-coding regulatory elements of HLA. The HLA region of the human genome accounts for the majority of risk in a number of autoimmune disorders. Our understanding of this risk is incomplete. This project aims at using genetic editing tools to investigate HLA variants in both the coding and non-coding region of the genome. Furthermore, we aim to develop methods for the targeted deletion of specific HLA alleles.
